Kang, 17, collected seven birdies against two bogeys at the Country Club of North Carolina to build a three-shot lead over three players.
She will try to win stroke-play medalist honors when the second round is contested Tuesday. The champion is crowned after six rounds of match play.
Kang was the stroke-play medalist at last year's U.S. Women's Amateur, but was knocked out in the first round of match play. She made the cut two weeks ago at the U.S. Women's Open and finished in 64th place.
Summar Roachell, Megan Khang and Ginger Howard share second place at two-under 70, while Jisoo Keel is the only player at 71.
Kristen Park, the 2007 champion, opened with a one-over 73 and is tied for ninth place. Park is the only former USGA winner in the field this week. She is trying to become just the fourth player in history to win this championship more than once.
